目的:为了制备高效的,特异的抗人喉癌/抗蓖麻毒素A双功能单克隆抗体,观察其对Hep-2细胞株杀伤作用。方法:选用BALB/C鼠,分别行蓖麻毒素A和人喉鳞状细胞癌腹腔免疫后,提取抗体,经与骨髓瘤细胞融合,制备成抗人喉癌/抗蓖麻毒素A双特异性单克隆抗体。结果:该双功能抗体与喉癌组织的特异性结合率为87%,对Hep-2细胞半数抑制浓度比游离环磷酰胺强40倍。结论:采用二次杂交瘤技术制备的双功能抗体优于化学偶联法,抗体效价具有均匀性和理想的信噪比,抗体易于标准化。
OBJECTIVE: To prepare a highly effective and specific anti-human laryngeal carcinoma / anti-ricin A dual-function monoclonal antibody and observe its killing effect on Hep-2 cell line. Methods: BALB / C mice were selected and immunized intraperitoneally with ricin A and human laryngeal squamous cell carcinoma respectively. Antibodies were extracted and fused with myeloma cells to prepare anti-human laryngeal carcinoma / anti-ricin A bispecific Monoclonal antibodies. Results: The specific binding rate of the bifunctional antibody to laryngeal carcinoma was 87%, and the inhibitory concentration of Hep-2 cells was 40 times stronger than that of free cyclophosphamide. Conclusion: The bifunctional antibody prepared by the second hybridoma technique is better than the chemical conjugation method. The antibody titer is uniform and the ideal signal-to-noise ratio, and the antibody is easy to standardize.
