目的 :研究冷应激对大鼠垂体前叶离体细胞ACTH分泌和POMC基因表达的影响及锌的保护作用 .方法 :采用原代培养的大鼠垂体前叶离体细胞 ,培养液中加入不同剂量的锌 ,并给予低温处理 .放射免疫法测定培养液中的ACTH ,斑点杂交法 (Dotblot)检测POMCmRNA水平 .结果 :一定浓度的锌对垂体前叶离体细胞ACTH的分泌有明显的促进作用 ;冷刺激后 ,垂体前叶细胞ACTH的分泌明显增加 ,加锌组ACTH增加的幅度有所降低 ;冷刺激使POMCmRNA的表达明显增强 ,补锌可使冷暴露大鼠垂体前叶POMCmRNA的表达水平有所下降 .结论 :锌对冷刺激引起的垂体前叶离体细胞ACTH分泌增加和POMC基因表达增强有一定的缓解作用
OBJECTIVE: To study the effect of cold stress on ACTH secretion and POMC gene expression in isolated rat anterior pituitary cells and the protective effect of zinc.Methods: Primary cultured rat anterior pituitary cells were cultured in vitro, Dose of zinc, and given low temperature treatment.The radioimmunoassay was used to determine the ACTH in culture medium and the dot blot assay to detect the level of POMC mRNA.Results: A certain concentration of zinc significantly promoted the secretion of ACTH in the anterior pituitary cells After cold stimulation, the secretion of ACTH in the anterior pituitary cells was significantly increased, while the increase of ACTH in the zinc group was reduced. The expression of POMC mRNA was significantly enhanced by cold stimulation. The level of POMC mRNA expression in the anterior pituitary of rats with cold exposure was increased by zinc supplementation Decreased.Conclusion: Zinc can alleviate the increase of ACTH secretion and POMC gene expression in isolated anterior pituitary cells induced by cold stimulation
