目的:研究长波紫外照射对血细胞中DNA的损害及对DNA检测造成的影响。方法:制备3种不同浓度的血样本,使用长波紫外光源照射不同时间后,再进行STR基因座分型检测,并对检测结果及基因座的缺失情况进行统计。结果:原血样本经紫外线照射前后都能得到完整的基因分型,少量血样本照射10 min后基因座减少,微量血样本照射5 min后基因座减少。长度为200 bp以上的STR位点较易缺失。结论:长波紫外照射会造成血液中DNA的损伤,原血样本不会影响DNA的检测,但低浓度的血样本,浓度越高,照射时间越长,STR分型检测中基因座缺失越多。
OBJECTIVE: To study the damage of DNA in blood cells by long-wave UV irradiation and its effect on DNA detection. Methods: Three kinds of blood samples with different concentrations were prepared. After being irradiated by long-wave ultraviolet light source for different time, STR genotypes were detected by typing, and the detection results and the deletion of loci were counted. Results: The complete genotyping of the original blood samples before and after UV irradiation could be obtained. The loci decreased after a small amount of blood samples were irradiated for 10 minutes, and the loci decreased after the blood samples were irradiated for 5 minutes. STR loci of more than 200 bp in length are more susceptible to deletion. Conclusion: Long-wave UV irradiation can cause DNA damage in the blood. The original blood sample does not affect the DNA detection. However, the low concentration of blood sample, the higher the concentration, the longer the irradiation time, the more genotype loss in the STR typing test.
