目的利用NO合酶抑制剂NG-硝基-L-精氨酸甲基酯盐酸盐(L-NAME)抑制胚胎细胞产生NO,探讨NO作为细胞功能活动的重要信使分子在早期胚胎发育中的调控作用。方法通过体外受精方式获得受精胚,将受精胚培养于添加不同浓度L-NAME的胚胎培养液中,检测胚胎发育的卵裂率、囊胚率和囊胚的总细胞数。结果 0.1mmol/L的L-NAME对胚胎的卵裂率和囊胚率没有显著影响,但囊胚的总细胞数显著降低;随着L-NAME浓度的增加,1和10 mmol/L L-NAME组胚胎的卵裂率、囊胚率和囊胚的总细胞数显著降低。结论 NO在早期胚胎发育过程中具有重要的调控作用,抑制胚胎细胞中NO的产生将严重影响早期胚胎的发育及发育质量。
OBJECTIVE: To inhibit the production of nitric oxide (NO) by NG-nitro-L-arginine methyl ester hydrochloride (L-NAME), an NO synthase inhibitor, and to explore the role of NO as an important messenger molecule in cell function during early embryo development Regulation. Methods The fertilized embryos were obtained by in vitro fertilization. The fertilized embryos were cultured in embryo medium supplemented with different concentrations of L-NAME to detect the cleavage rate, blastocyst rate and total blastocyst count of embryos. Results L-NAME with 0.1 mmol / L had no significant effect on the cleavage rate and blastocyst rate of embryos, but the total number of blastocysts decreased significantly. With the increase of L-NAME concentration, 1 and 10 mmol / L L- The cleavage rate, blastocyst rate and total blastocyst count of NAME group embryos were significantly decreased. Conclusions NO plays an important regulatory role in early embryonic development. Inhibition of NO production in embryonic cells will seriously affect the development and developmental quality of early embryos.
