目的 :探讨FasL基因在体内诱导胶质瘤细胞凋亡和抑制增殖的作用。方法 :在裸鼠皮下种植Fas表达阳性的人脑胶质母细胞瘤细胞系TJ90 5 ,成瘤后 ,利用脂质体介导FasL基因瘤内治疗 ,在 3 9d观察期间 ,用千分尺测量肿瘤生长 ,最终取出肿瘤 ,用原位杂交 ,免疫组化鉴定FasL基因和蛋白的表达 ,应用TUNEL法检测细胞凋亡 ,Ki 67免疫组化检测细胞增殖。结果 :FasL基因瘤内注射观察 3 9d后 ,治疗组瘤体积明显小于对照组 (P <0 0 1) ,肿瘤组织细胞内FasL基因表达增加 ,凋亡细胞数增多 ,细胞增殖率降低。结论 :FasL基因对Fas表达阳性胶质瘤有治疗作用。
Objective: To investigate the role of FasL gene in inducing glioma cell apoptosis and inhibiting proliferation in vivo. METHODS: Fas-positive human brain glioblastoma cell line TJ905 was implanted subcutaneously in nude mice. After tumorigenesis, the tumor was treated with liposomes mediated by FasL gene. The tumor growth was measured by micrometer during 39 days observation The tumor was finally removed and the expression of FasL gene and protein was identified by in situ hybridization and immunohistochemistry. The apoptosis was detected by TUNEL method and the proliferation of cells was detected by immunohistochemistry. Results: The tumor volume of the treated group was significantly smaller than that of the control group (P <0.01) after 39 days of intratumoral injection of FasL gene. The expression of FasL gene in the tumor tissue increased, the number of apoptotic cells increased and the cell proliferation rate decreased. Conclusion: FasL gene has a therapeutic effect on Fas-positive glioma.
