为了确定毛细胞白血病(HCL)细胞的存活与生长因子的关系,作者在体外研究了肿瘤坏死因子(TNF)、IL—1α、IL-1β、IL—2、IL—3、IL—4、IL—5、IL—6、B细胞生长因子(BCGF)、GM—CSF、PHA—LCM、5637膀胱肉瘤条件培养液等对HCL细胞的效应。HCL细胞来自5例HCL患者的脾脏(HC25、HC56、HC67、HC58、HC59),所有患者均未进行化疗。脾脏取出后制备成单个核细胞,并行TRAP染色及细胞免疫表型分析,以正常脾细胞作为对照。细胞以3.33×10~6/ml的浓度种入96孔培养板,培养体系为1MDM+8%FBS+8%HS+2mM谷氨酰胺+0.22mM肌醇,培养条件为5%CO_2培养箱,分33℃和37℃两种温度条件。
To determine the relationship between survival and growth factors in hairy cell leukemia (HCL) cells, the authors studied tumor necrosis factor (TNF), IL-1 alpha, IL-1 beta, IL-2, IL-3, IL-4, IL in vitro. — 5, IL-6, B-cell growth factor (BCGF), GM-CSF, PHA-LCM, 5637 bladder sarcoma conditioned medium and other effects on HCL cells. HCL cells were derived from the spleen of 5 patients with HCL (HC25, HC56, HC67, HC58, HC59) and none of the patients were treated with chemotherapy. After the spleen was removed, mononuclear cells were prepared and stained with TRAP and immunophenotype analysis. Normal splenocytes were used as controls. The cells were seeded into a 96-well culture plate at a concentration of 3.33×10 6 /ml. The culture system was 1MDM+8%FBS+8%HS+2mM glutamine+0.22mM inositol, and the culture condition was a 5% CO 2 incubator. Divided into 33 °C and 37 °C two temperature conditions.
