目的:建立高效液相色谱-质谱法同时测定菟丝子药材中5种成分的含量。方法:采用Agilent ZorbaxSB-C18(2.1 mm×100 mm,3.5μm)色谱柱;流动相:0.05%甲酸水溶液-乙睛,梯度洗脱;检测波长:360 nm;流速:0.4 mL/min;柱温:45℃。Finnigan电喷雾离子阱多级质谱仪;负离子检测模式;ESI喷雾电压:4.5 kV;鞘气(N2)流速:60 a.u.;辅助气(N2)流速:20 a.u.;毛细管温度:300℃;毛细管电压:-19 V,扫描范围m/z:90~800。结果:色谱峰分离情况良好,菟丝子中5种化学成分在各自的浓度范围内均具有良好的线性关系;加样回收率为96.06%~101.61%,RSD为0.67%~2.90%。结论:所建方法简便、准确,重现性好,可用于菟丝子药材的质量评价。 Objective: To establish a HPLC-MS method for the simultaneous determination of five components in Dodder. The mobile phase consisted of 0.05% formic acid aqueous solution - acetonitrile and gradient elution. The detection wavelength was 360 nm and the flow rate was 0.4 mL / min. The column temperature : 45 ° C. Finnigan electrospray ion trap mass spectrometer; negative ion detection mode; ESI spray voltage: 4.5 kV; gas flow rate of sheath gas: 60 au; flow rate of auxiliary gas: 20 au; capillary temperature: -19 V, scanning range m / z: 90 ~ 800. Results: The separation of chromatographic peaks was good. The five chemical components of Dodder had a good linear relationship in their concentration range. The recoveries were 96.06% -101.61% with RSD 0.67% -2.90%. Conclusion: The method is simple, accurate and reproducible, which can be used to evaluate the quality of Dodder.
