目的研究宫颈上皮内瘤变及宫颈癌组织中人乳头状瘤病毒(HPV)感染与抑癌基因P16表达的关系。方法采用免疫组织化学法检测35例宫颈癌、45例子宫上皮内瘤变(CIN)和30例慢性宫颈炎组织的P16基因的表达情况,同时采用PCR-反向杂交法对其宫颈细胞进行HPV分型检测。结果从慢性宫颈炎、CIN到宫颈癌,各组病例HPV的感染率和P16基因的表达率均呈递增趋势,CIN组和宫颈癌组HPV感染率和P16基因的表达率显著高于慢性宫颈炎组,差异有统计学意义(P<0.01)。在CIN与宫颈癌病例组织中,P16基因的表达率与HPV感染率呈正相关(r=0.98,P<0.05)。结论在CIN、宫颈癌发生发展过程中,HPV感染和P16基因表达均发挥重要作用,而且HPV感染是导致P16基因高表达的原因之一。
Objective To investigate the relationship between human papillomavirus (HPV) infection and the expression of tumor suppressor gene P16 in cervical intraepithelial neoplasia and cervical cancer. Methods Immunohistochemical method was used to detect the expression of P16 gene in 35 cases of cervical cancer, 45 cases of uterine intraepithelial neoplasia (CIN) and 30 cases of chronic cervicitis. Meanwhile, HPV16 positive cells were detected by PCR-reverse hybridization Typing test. Results From chronic cervicitis and cervical intraepithelial neoplasia, HPV infection rate and P16 gene expression rate showed an increasing trend in each group, HPV infection rate and P16 gene expression rate in CIN group and cervical cancer group were significantly higher than those in chronic cervicitis Group, the difference was statistically significant (P <0.01). In CIN and cervical cancer cases, the expression of P16 was positively correlated with HPV infection (r = 0.98, P <0.05). Conclusion HPV infection and P16 gene expression play an important role in CIN and cervical cancer development, and HPV infection is one of the reasons leading to the high expression of P16 gene.
