本文用DPH~(**)荧光探剂研究Mg~(2+)、Ca~(2+)、Mn~(2+)、Sr~(2+)、Zn~(2+)、Cd~(2+)等六种金属离子及胆固醇对脂质体或嵌有猪心线粒体H~+—ATP酶的脂酶体—L·(H~+—ATP酶)流动性的影响,并比较了上述因素影响重建H~+—ATP酶活性的作用。通过胆酸盐透析法形成结合有二价金属离子的脂质体或嵌有猪心线粒体H~+—ATP酶的脂酶体。研究表明Mg~(2+)、Ca~(2+)、Mn~(2+)、Sr~(2+)、Zn~(2+)、Cd~(2+)等六种二价金属离子,均有使脂质体或L·(H~+—ATP酶)的流动性降低的作用。但是,Mg~(2+)、Ca~(2+)、Mn~(2+)、Sr~(2+)具有提高重建H~+—ATP酶活性的作用,而Zn~(2+)和Cd~(2+)却反而降低重建H~+—ATP酶活性。提高重建H~+—ATP酶对寡霉素的敏感性的顺序为Mg~(2+)>Ca~(2+)>Mn~(2+)>Sr~(2+)。11%胆固醇使L·(H~+—ATP酶)流动性降低程度几乎与ImM Mg~(2+)的作用相等,然而,后者提高重建H~+—ATP酶活性及其对寡霉素的敏感性一倍左右,前者却降低近25%。因此,Mg~(2+)提高重建猪心线粒体H~+—ATP酶的活性似乎具有一定的专一性。
In this paper, the effects of Mg 2+, Ca 2+, Mn 2+, Sr 2+, Zn 2+, Cd ~ (-) 2+) and cholesterol on the fluidity of liposomes or H + -ATPase-loaded lipase-L · (H ~ + -ATPase) Factors Affecting the Reconstitution of H ~ + -ATPase Activity. Liposomes incorporating bivalent metal ions or H + -ATPase-laden liposomes embedded with porcine heart mitochondria were formed by cholate dialysis. The results show that six kinds of divalent metal ions such as Mg2 +, Ca2 +, Mn2 +, Sr2 +, Zn2 +, Cd2 + , Both of which reduce the fluidity of liposomes or L · (H ~ + -ATPase). However, Mg2 +, Ca2 +, Mn2 + and Sr2 + could enhance the activity of H + -ATPase, while Zn2 + and Mn2 + Cd ~ (2+) can reduce the rebuilding H ~ + -ATPase activity instead. The order of improving the sensitivity of H ~ + -ATPase to oligomycin was Mg ~ (2 +)> Ca ~ (2 +)> Mn ~ (2 +)> Sr ~ (2+). 11% cholesterol reduced the fluidity of L · (H ~ + -ATPase) almost equal to that of ImM Mg ~ (2+), however, the latter increased the activity of reconstituted H ~ + -ATPase and its effect on oligomycin About double the sensitivity, the former has dropped nearly 25%. Therefore, Mg ~ (2+) seems to have a certain specificity to improve the activity of mitochondrial H ~ + -ATPase in porcine heart.
