目的探讨脂质体介导的碱性成纤维细胞生长因子(bFGF)mRNA的反义寡核苷酸(ASODN)对大鼠前列腺组织的影响。方法将45只SD大鼠随机分为反义组、正义组和对照组,向大鼠前列腺组织中分别注入人工合成的bFGFmRNA的反义寡核苷酸脂质体,正义寡核苷酸脂质体和无药脂质体,分别在术后第4、7、14天取出大鼠前列腺,测大鼠前列腺重量及前列腺指数,逆转录聚合酶链反应(RTPCR)检测大鼠前列腺组织中bFGFmRNA的表达,免疫组织化学染色结合计算机图像分析对大鼠前列腺组织中bFGF和增殖细胞核抗原(PCNA)表达进行定量分析。结果将拟给药物注入大鼠前列腺内后7d,正义组大鼠前列腺指数(2.03±0.17)和对照组大鼠前列腺指数(2.02±0.18)相比差异无统计学意义(P>0.05),但均明显高于反义组大鼠前列腺指数(1.62±0.05,P<0.01),正义组bFGFmRNA表达水平(0.5894±0.0245)和对照组bFGFmRNA表达水平(0.6442±0.0569)相比差异无统计学意义(P>0.05),但均明显高于反义组bFGFmRNA表达水平(0.2658±0.0337,P<0.01)。正义组和对照组大鼠前列腺组织中bFGF表达和PCNA指数相比差异无统计学意义(P>0.05),而反义组大鼠前列腺组织中bFGF表达和PCNA指数则显著低于正义组和对照组(P<0.01),且有随时间推移而逐渐下降的趋势。结论脂质体转染bFGFmRNAASODN对
Objective To investigate the effect of liposome-mediated basic fibroblast growth factor (bFGF) antisense oligonucleotide (ASODN) on rat prostate tissue. Methods 45 Sprague-Dawley rats were randomly divided into antisense group, sense group and control group. Antisense oligonucleotide liposomes of synthetic bFGF mRNA were injected into rat prostate tissue respectively. The rats were sacrificed on the 4th, 7th and 14th days after surgery respectively. Prostate weight and prostate index were measured. Reverse transcriptase polymerase chain reaction (RTPCR) was used to detect the expression of bFGF mRNA Expression, immunohistochemical staining combined with computer image analysis of prostate tissue bFGF and PCNA expression quantitative analysis. Results There was no significant difference in prostate index (2.03 ± 0.17) and prostate index (2.02 ± 0.18) between the rats in the control group and the control group on the 7th day after injection of the drugs into the prostate of rats, but there was no significant difference between the two groups (P> 0.05) (1.62 ± 0.05, P <0.01). There was no significant difference in the expression of bFGF mRNA (0.5894 ± 0.0245) and bFGF mRNA (0.6442 ± 0.0569) in the control group P> 0.05), but both of them were significantly higher than that of antisense group (0.2658 ± 0.0337, P <0.01). There was no significant difference in the expression of bFGF and PCNA between the positive group and the control group (P> 0.05), while the expression of bFGF and PCNA in the prostate tissue of the antisense group was significantly lower than that of the normal group and the control group Group (P <0.01), and there was a tendency to decline gradually over time.
