目的采用Q-TRAP LC-MS/MS测定杏苏散水提液中活性成分苦杏仁苷、芦丁、木犀草素和白花前胡乙素的含量。方法采用Supelco Discovery C-18(3μm,2.1 mm×150 mm)色谱柱,以乙腈和水(含体积比0.1%甲酸)为流动相,梯度洗脱,流速为0.25 ml/min,AB Sciex 4000 Q-TRAP型质谱仪多重反应监测(MRM)扫描模式对杏苏散水提液样品进行检测。结果杏苏散水提液中苦杏仁苷、芦丁、木犀草素和白花前胡乙素的含量分别为63.18μg/ml、0.47μg/ml、1.65μg/ml和0.13μg/ml。结论该方法简便、准确、重现性好,可用于对杏苏散水提液中苦杏仁苷、芦丁、木犀草素和白花前胡乙素这4种成分进行定量分析。
OBJECTIVE To determine the content of amygdalin, rutin, luteolin and bcl-2 in water extract of Xingsu Powder by Q-TRAP LC-MS / MS. Methods A gradient elution was performed on a Supelco Discovery C-18 column (3 μm, 2.1 mm × 150 mm) using a mobile phase of acetonitrile and water (0.1% formic acid) with a flow rate of 0.25 ml / min and AB Sciex 4000 Q -TRAP mass spectrometer multiple reaction monitoring (MRM) scanning mode of the samples were tested for water samples. Results The content of amygdalin, rutin, luteolin and pre-apocynin in the aqueous extract of Xingsu San were 63.18μg / ml, 0.47μg / ml, 1.65μg / ml and 0.13μg / ml respectively. Conclusion The method is simple, accurate and reproducible. It can be used to quantitatively analyze the four components of amygdalin, rutin, luteolin and presenilin in the aqueous extract of Xingsu San.
