目的探讨P53负调控蛋白Apak的组织分布特征并分析其白血病相关性。方法利用组织斑点杂交分析Apak在多种组织中的分布情况,Northern杂交分析其在5种白血病细胞系的转录,并用RT-PCR分析Apak在32例白血病患者和12例健康人外周血中的mRNA水平。结果 Apak的组织表达谱分析表明该基因只在胎肝、胎脾以及成人唾液腺、膀胱、左右心室和心尖中高表达。在多种白血病细胞系Jurkat,AHH-1,MOLT-4,SKO-007和HL-60和部分白血病患者的外周血中Apak也呈现高转录,提示Apak在白血病发生中可能发挥重要功能。结论本研究揭示了Apak是一种具有组织特异性分布的P53负调节蛋白,其在白血病中的高表达提示其可能与该病的发生有一定的关联。
Objective To investigate the tissue distribution of P53, a negative regulator of Apak, and analyze its association with leukemia. Methods The distribution of Apak in various tissues was analyzed by dot blot hybridization. Northern blotting was used to analyze the expression of Apak in five leukemia cell lines. The mRNA expression of Apak in peripheral blood of 32 leukemia patients and 12 healthy volunteers was analyzed by RT-PCR. Level. Results Apak tissue expression profiling showed that the gene was highly expressed only in fetal liver, fetal spleen and adult salivary glands, bladder, left ventricle, and apex. Apak also showed high transcription in the peripheral blood of a variety of leukemia cell lines Jurkat, AHH-1, MOLT-4, SKO-007 and HL-60 and some leukemia patients, suggesting that Apak may play an important role in the development of leukemia. Conclusion This study revealed that Apak is a tissue-specific distribution of P53 negative regulatory protein, and its high expression in leukemia suggests that it may be associated with the occurrence of the disease.
