目的探讨5-氟胞嘧啶(5-FC)/CD∷UPRT联合基因系统对大鼠脑胶质瘤的治疗作用。方法以逆转录病毒介导构建C6-CD∷UPRT细胞系,应用立体定向技术移植C6-CD∷UPRT细胞至SD大鼠尾状核,设立C6-pLXSN和C6细胞为对照组,建立脑胶质瘤荷瘤鼠模型,应用5-FC腹腔注射治疗,观察肿瘤大小、大鼠存活期、电镜观察细胞形态和流式细胞术检测肿瘤细胞的凋亡指标的变化。结果5-FC/CD∷UPRT治疗组(A组)与各对照组相比,A组肿瘤直径平均为(4.2±0.7)mm,远小于对照组的(8.3±1.3)mm,(P<0.01);对照组平均存活期(32±3)d,A组为(85±2)d(P<0.01);A组电镜显示核染色质分裂明显,并见典型的凋亡小体;A组肿瘤细胞中凋亡细胞所占百分率为19.36%。结论5-FC/CD∷UPRT联合基因对大鼠脑胶质瘤有显著的治疗效果。
Objective To investigate the therapeutic effect of 5-fluorocytosine (5-FC) / CD :: UPRT combined gene system on glioma in rats. Methods C6-CD :: UPRT cell line was constructed by retrovirus. C6-CD :: UPRT cells were transplanted to caudate nucleus of SD rats by stereotactic technique. C6-pLXSN and C6 cells were established as control group to establish glial The tumor-bearing mice model was treated with 5-FC intraperitoneal injection. The tumor size, the survival time of the rats, the morphology of the tumor were observed by electron microscope and the apoptosis index of the tumor cells were detected by flow cytometry. Results The tumor diameter in group A was (4.2 ± 0.7) mm, much smaller than that in control group (8.3 ± 1.3) mm, P <0.01 ). The average survival time in the control group was (32 ± 3) days, and in the A group was (85 ± 2) days (P <0.01). The electron microscopy results showed that the chromatin split was obvious in group A, The percentage of apoptotic cells in tumor cells was 19.36%. Conclusions 5-FC / CD :: UPRT combined gene has a significant therapeutic effect on glioma in rats.
