目的探讨人非小细胞肺癌细胞A549中孕酮对生长抑素受体(somatostatinrecepter,SSTR)表达的调节。方法免疫组化法测定A549细胞孕激素受体的表达,RT-PCR检测孕酮作用前后SSTR亚型mRNA的表达。结果A549细胞表达孕激素受体,同时表达生长抑素受体亚型SSTR2、SSTR1、SSTR4和SSTR5mRNA,不表达SSTR3mRNA。不同浓度的孕酮作用A549细胞24h后,SSTR各亚型mRNA表达普遍上调(P<0.05);10nmol/L组及1000nmol/L组细胞出现了SSTR3mRNA的表达。延长10nmol/L孕酮作用时间至48h,SSTR3与SSTR5mRNA表达继续上调(P<0.05)。结论孕酮上调A549细胞SSTR各亚型mRNA的表达,尤其是SSTR3与SSTR5mRNA;调节作用与孕酮的作用时间和浓度有关。
Objective To investigate the regulation of somatostatin receptor (somatostatin receptor) (SSTR) expression in human non-small cell lung cancer A549 cells. Methods The expression of progesterone receptor in A549 cells was detected by immunohistochemistry. The mRNA expression of SSTR was detected by RT-PCR. Results A549 cells expressed progesterone receptor and expressed somatostatin receptor subtypes SSTR2, SSTR1, SSTR4 and SSTR5 mRNA, but not SSTR3 mRNA. After treated with different concentrations of progesterone for 24 hours, the mRNA expression of SSTR subtypes was generally up-regulated (P <0.05). The expression of SSTR3 mRNA appeared in 10 nmol / L and 1000 nmol / L groups. Prolonged 10nmol / L progesterone treatment time to 48h, SSTR3 and SSTR5mRNA expression continued to increase (P <0.05). Conclusion Progesterone up-regulates the expression of SSTR subtypes in A549 cells, especially SSTR3 and SSTR5 mRNA. The regulative effect of progesterone on the expression of progesterone is related to the time and concentration of progesterone.
