目的:研究硫磺熏蒸对怀牛膝中蜕皮甾酮的影响。方法:采用高效液相色谱法测定硫磺熏蒸后蜕皮甾酮的含量,XB-C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-水(15:85),检测波长250 nm,柱温25℃,流速0.8 mL.min-1。结果:硫磺用量100 g.m-3,熏蒸1次,熏蒸2 h,此时蜕皮甾酮的量最高;蜕皮甾酮在0.408~2.448μg线性关系良好,回归方程为Y=34.167X+61(r=0.999 6)。结论:硫磺熏蒸对怀牛膝中蜕皮甾酮含量有一定影响,随硫磺用量增加、熏蒸时间延长、熏蒸次数增加而呈减少趋势。
Objective: To study the effect of sulfur fumigation on ecdysterone in Achyranthes bidentata. Methods: The content of ecdysterone after sulfur fumigation was determined by high performance liquid chromatography (HPLC). The chromatographic column of XB-C18 (4.6 mm × 250 mm, 5 μm), mobile phase of acetonitrile- water (15:85) Temperature 25 ℃, flow rate 0.8 mL.min-1. The results showed that the amount of ecdysterone was the highest at 100 gm-3 and fumigation for 2 h. The linear relationship of ecdysterone was 0.408 ~ 2.448μg, the regression equation was Y = 34.167X + 61 (r = 0.999 6). Conclusion: Sulfur fumigation has a certain influence on the ecdysterone content in Achyranthes bidentata. With the increase of the amount of sulfur, the fumigation time is prolonged and the number of fumigation is increasing, showing a decreasing trend.
