目的:观察肾康注射液对高血压肾损害大鼠血压、代谢、血液生化、肾脏组织病理学等的影响,为临床试验提供理论依据。方法:将75只高血压肾损伤SHR随机分为5组:模型组、苯那普利阳性对照组及肾康注射液低、中、高剂量组,每组15只,以WKY为正常对照组(n=15)。低、中、高剂量组分别给予肾康注射液6.7、13.3、26.6m L/kg,相当于生药2、4、8 g/kg。模型组和正常对照组给予等体积生理盐水,苯那普利组给药剂量8 mg/kg。给药方式为腹腔注射。结果:连续腹腔注射给药3个月,肾康注射液能抑制SHR大鼠血压升高;显著降低SHR尿液中尿微量白蛋白及尿蛋白浓度;降低血肌酐(Scr)和尿素氮(BUN)含量;提高内生肌酐清除率(Ccr)和血清白蛋白(ALB)含量及超氧化物歧化酶(SOD)活性,降低血清中丙二醛(MDA)、醛固酮(Ald)、肿瘤坏死因子(TNF-α)、白介素(IL-6)、转化生长因子(TGF-β1)和C反应蛋白(CRP)的含量;各组大鼠肾脏指数与心脏系数差异均无统计学意义,肾脏病理切片显示给药后肾小球纤维化明显降低,炎细胞减少,肾小管纤维化减轻。结论:肾康注射液具有改善高血压肾损害大鼠肾脏功能、降低血清炎症因子水平、改善氧化应激状态及减轻高血压肾损害程度等作用。
Objective: To observe the effects of Shenkang injection on blood pressure, metabolism, blood biochemistry and renal histopathology in rats with hypertensive renal failure, and to provide a theoretical basis for clinical trials. Methods: SHRs of 75 hypertensive renal injuries were randomly divided into five groups: model group, benazepril positive control group and Shenkang injection low, medium and high dose group, 15 rats in each group. There was no significant difference in renal index and cardiac coefficient between groups After administration of glomerular fibrosis was significantly reduced inflammatory cells decreased tubular fibrosis alleviated. Conclusion: Shenkang injection can improve renal function, reduce serum levels of inflammatory cytokines, improve oxidative stress status and reduce the degree of renal damage in hypertensive rats with renal damage.
