目的:观察清热化瘀颗粒对脑缺血预处理大鼠缺血再灌注后肌醇需求激酶1(IRE1)mRNA及其蛋白表达的影响。方法:健康雄性SD大鼠80只,随机分为4组:假手术组(SO)、脑缺血再灌注组(MCAO)、脑缺血预处理组(BIP)、清热化瘀颗粒组(QRHY),每组按再灌注后12 h,1,2,3 d 4个时间点平均分为4个亚组。制备脑缺血预处理模型,缺血预处理后给大鼠灌胃14 mL·kg-1清热化瘀颗粒溶液,连续3 d,每天上、下午各1次,MCAO组和BIP组给予等量的生理盐水。用实时荧光定量PCR和Western blot法观察再缺血后各个时间点IRE1mRNA及其蛋白的表达变化。结果:MCAO组IRE1mRNA及其蛋白表达均于缺血再灌注后12 h开始明显上升,1 d达高峰(P<0.01),随再灌注时间延长其表达逐渐下降,但仍保持较高表达水平(P<0.01);BIP组较MCAO组IRE1mRNA及其蛋白表达明显升高(P<0.05或P<0.01),QRHY组较BIP组进一步升高其表达(P<0.05或P<0.01)。结论:脑缺血预处理能够诱导脑缺血损伤后IRE1的表达,清热化瘀颗粒可进一步促进其表达。
Objective: To observe the effect of Qingre Huayu Granule on IRE1 mRNA and protein expression after cerebral ischemic preconditioning in rats. Methods: Eighty healthy male Sprague Dawley rats were randomly divided into 4 groups: sham operation group (SO), cerebral ischemia reperfusion group (MCAO), cerebral ischemic preconditioning group (BIP) ). Each group was divided into 4 subgroups at 4 h, 12 h, 1, 2 and 3 d after reperfusion. The model of cerebral ischemic preconditioning was established. After the ischemic preconditioning, rats were given 14 mL · kg-1 Qingre Huayu Granule solution for 3 consecutive days, once a day and once a day. The rats in MCAO group and BIP group were given the same dose Saline. Real-time quantitative PCR and Western blot were used to observe the expression of IRE1 mRNA and protein at different time points after ischemia. Results: The expression of IRE1 mRNA and its protein in MCAO group both increased significantly at 12 h after ischemia-reperfusion and peaked on the 1st day (P <0.01). The expression of IRE1 mRNA decreased gradually with the reperfusion time, but remained high P <0.01). The expression of IRE1 mRNA and protein in BIP group was significantly higher than that in MCAO group (P <0.05 or P <0.01). The expression of IRE1 in QRIP group was higher than that in BIP group (P <0.05 or P <0.01). Conclusion: Cerebral ischemia preconditioning can induce the expression of IRE1 after cerebral ischemic injury, and Qingre Huayu granules can further promote its expression.
