已证实抑制生长素((Somatostatin)生长激素释放抑制因子)能有效地控制急性静脉曲张出血。然而,抑制生长素的生物半衰期非常短,需要静脉输入以维持所需的血浓度。近来,已生产了一种抑制生长素类似物SMS201-995,它的作用时间长,可以克服上述一些缺点。为了阐明SMS 201-995的作用机理,作者检测其在猪肝脏中的血液动力学中的作用,并确定其是否降低肝硬变和门脉高压病人的曲张静脉内压。 (一)动物实验取母猪在全麻下剖腹,6只猪静脉滴入SMS201-995250μg/h,共20秒钟,总量为6 ml。4只猪输入等量的等渗盐水作对照,连续记录门脉压、门静脉流量、肝动脉血流、动脉压和脉率,由此分别计算出内脏静脉和门脉阻力。
Inhibition of somatostatin (somatostatin) growth inhibitory factor has been shown to be effective in controlling acute variceal bleeding. (A) Animal Experiments Sows were sowed under general anesthesia and 6 pigs were intravenously administered with SMS201-995250 μg / h for 20 seconds in a total amount of 6 ml. Four pigs were given the same amount of isotonic saline as a control, and portal pressure, portal vein flow, hepatic artery blood flow, arterial pressure and pulse rate were continuously recorded, thereby calculating visceral and portal vein resistance.
