1978年我们在研究国内外优良培养基的各种成分配比的基础上,对培养基中5种大量元素进行了筛选。采用了5种元素4种水平的正交设计法试验[L_(16)(4~5)]。经过两年来的6次重复,41个处理的多次试验,筛选出一种适于花药培养的优良培养基,定名为“正14”。 1978年在正交试验的16种培养基上,接种玉米品种海淀八趟白,其结果以“正14”培养基的诱导频率最高,在试验的所有培养基中占首位。 1979年用“正14”培养基对玉米单三91,九单1×京黄13等11份材料进行试验,其平均诱导率为15.4%,绿苗诱导率为2.1%,而对照培养基N_6的愈伤组织诱导率和绿苗诱导率分别为5.4%和1.3%,
In 1978, we studied five kinds of macronutrients in culture medium on the basis of studying the ratio of various components of excellent medium at home and abroad. Orthogonal design test of four levels of five elements was used [L_ (16) (4 ~ 5)]. After six replications and 41 treatments over the past two years, a good medium suitable for anther culture was screened and named as “positive 14”. In 1978, 16 maize cultivars were inoculated with maize variety Haishu 8 times in orthogonal experiment. The result showed that the induction frequency was the highest in “14” medium and the highest in all medium tested. In 1979, “Zheng 14” medium was used to test 11 materials such as corn single 91, 9 single 1 × Jinghuang 13, the average induction rate was 15.4% and the green plantlet induction rate was 2.1%, while the control medium N_6 The callus induction rate and green shoot induction rate were 5.4% and 1.3% respectively,
