目的:建立了测定组织中的丝裂霉素C的反相高效液相色谱法。方法:组织匀浆用乙酸乙酯提取。以Shim-Pack CLC-ODS(150mm×6mm,10pm)为色谱柱,甲醇-水(35:65)为流动相,检测波长为365nm,外标法定量。结果:丝裂霉素C组组织浓度线性范围为0.1~2.0pg/g(r=0.9991,方法回收率为97.10%。日内、日间RSD%为4.00%~8.42%,最低检测浓度为0.05eg/g。结论:该法简便,适用于丝裂霉素C组织浓度的测定
OBJECTIVE: To establish an RP-HPLC method for the determination of mitomycin C in tissues. Methods: Tissue homogenates were extracted with ethyl acetate. The mobile phase consisted of Shim-Pack CLC-ODS (150mm × 6mm, 10pm) and methanol-water (35:65). The detection wavelength was 365nm. Results: The linear range of mitomycin C tissue concentration was 0.1-2.0 pg / g (r = 0.9991, the method recovery was 97.10%. The intra-day and inter-day RSD% was 4.00% -8.42%, the lowest detection concentration was 0.05eg / g. Conclusion: This method is simple, suitable for the determination of mitomycin C tissue concentration
