目的观察咳喘宁对RSV诱发哮喘大鼠血清IL-4、IFN-γ的调节作用及对气道平滑肌细胞(airway smooth muscle cells,ASMCs)增殖水平(OD值)的影响。方法将60只大鼠随机分为6组,除正常组外,其余大鼠通过致敏、诱喘、病毒激发哮喘进行造模。造模完成当天分组干预,并用ELISA进行IL-4、IFN-γ水平的测定。造模成功后取支气管平滑肌进行细胞培养,倒置显微镜下化学荧光法鉴定为平滑肌细胞。制备大鼠不同组含药血清并加入第3-7代ASMCs中干预。运用MTT法检测各组ASMCs增殖水平(OD值)。结果 ELISA法结果 ,与模型组相比,西药治疗组、咳喘宁各剂量组血清IL-4、IFN-γ水平差异具有统计学意义(P<0.05);与咳喘宁大、小剂量组相比,中剂量组血清IL-4、IFN-γ水平差异有统计学意义(P<0.05)。MTT法结果,与空白对照组比较,其余5组差异均有显著统计学意义(P<0.01);与咳喘宁大、小剂量血清中比较,中剂量血清组差异有统计学意义(P<0.05)。结论咳喘宁对治疗RSV诱发的哮喘大鼠疗效较为显著,通过上调IFN-r水平、降低IL-4水平表达从而降低气道炎症反应,维持Th1/Th2平衡来调节机体免疫机制。咳喘宁治疗支气管哮喘可能与其能够抑制气道平滑肌增殖、影响哮喘气道重塑关系密切。
Objective To observe the effect of Kechuanning on the regulation of serum IL-4 and IFN-γ in RSV-induced asthmatic rats and the proliferation of airway smooth muscle cells (ASMCs). Methods Sixty rats were randomly divided into six groups. Except normal group, the other rats were induced by sensitization, induction of asthma and virus-induced asthma. On the day of modeling, the interventions were divided into groups, and the levels of IL-4 and IFN-γ were measured by ELISA. Bronchial smooth muscle cells were obtained after successful modeling, and were identified as smooth muscle cells by inverted fluorescence microscope. Different groups of rat serum were prepared and added to the 3-7 generation ASMCs for intervention. MTT assay was used to detect the proliferation of ASMCs in each group (OD value). Results Compared with the model group, the levels of IL-4 and IFN-γ in serum of western medicine group and Kechuanning group were significantly different (P <0.05) by ELISA. Compared with the model group, Compared with the middle dose group, the serum levels of IL-4 and IFN-γ were significantly different (P <0.05). Compared with the blank control group, the other 5 groups showed significant difference (P <0.01) by MTT assay. Compared with the large and small dose Kehuang Ning serum group, the difference was statistically significant (P < 0.05). Conclusion Kechuanning treatment of RSV-induced asthma in rats more significant effect, by upregulating the level of IFN-r, reducing the expression of IL-4 to reduce airway inflammatory response and maintain Th1 / Th2 balance to regulate the body’s immune mechanism. Kechuanning treatment of bronchial asthma may be related to its ability to inhibit airway smooth muscle proliferation, affecting airway remodeling of asthma closely.
