目的:通过观察大豆苷元对前列腺癌细胞(PC-3)增殖和相关基因表达的影响,探讨通过膳食干预途径预防前列腺癌发病的可行性。方法:将PC-3在F12培养基(含10%小牛血清)中采用开放式单层贴壁培养,实验设溶剂对照组、受试物各六个剂量组,采用MTT法对PC-3的增殖情况进行分析,对大豆苷元作用后的雄激素非依赖型前列腺癌(PC-3)细胞的存活率、细胞毒作用进行了研究。结果:与溶剂对照组相比,大豆苷元对PC-3细胞抑制效应均具有剂量依赖性,具有诱导凋亡和引起坏死效应,同时能诱导PC-3细胞的(PTEN基因)表达。结论:大豆苷元具有明显抑制前列腺癌细胞PC-3增殖效应,并呈现剂量-效应关系。
Objective: To observe the effect of daidzein on the proliferation of prostate cancer cells (PC-3) and the expression of related genes, and to explore the feasibility of preventing the occurrence of prostate cancer through dietary intervention. Methods: PC-3 was cultured in open monolayer in F12 medium containing 10% fetal bovine serum. The experimental groups were divided into six groups: solvent control group and test group. PC-3 The effects of daidzein on the survival and cytotoxicity of androgen-independent prostate cancer (PC-3) cells were studied. Results: Compared with the solvent control group, daidzein inhibited the PC-3 cells in a dose-dependent manner, inducing apoptosis and inducing necrosis, and inducing the expression of PTEN gene in PC-3 cells. Conclusion: Daidzein can significantly inhibit the proliferation of PC-3 cells in a dose-dependent manner.
