为探讨尾吊大鼠骨骼肌线粒体Ca2+,Mg2+离子含量、体视学变化与肌肉功能降低的关系,采用SD雄性大鼠50只,体重180~220g,按体重配对随机分为对照组和尾吊组。尾吊组30只,-25°;对照组20只。尾吊第15d(15只),第30d(15只)断头,取比目鱼肌、腓肠肌冷冻待测试。采用差速离心分离线粒体,按Lowry方法[1]测线粒体蛋白,钙、镁含量。按北京医科大学面分析方法测线粒体体视学各指标[2]。尾吊15、30d大鼠比目鱼肌、腓肠肌线粒体Ca、Mg含量与对照比增加,体视学各指标均有不同程度的改变。结果表明,尾吊后大鼠骨骼肌线粒体Ca、Mg离子代谢和体视学均发生改变,其变化必然影响肌肉收缩功能。
In order to investigate the relationship between mitochondrial Ca2 +, Mg2 + ion content in skeletal muscle and the decrease of muscle function in skeletal muscle of tail-suspended rats, 50 male Sprague-Dawley rats weighing 180-220 g were randomly divided into control group and tail-suspension group. Tail hanging group 30, -25 °; control group of 20. Tail hanging 15d (15), 30d (15) decapitated, take soleus, gastrocnemius to be frozen. Mitochondria were isolated by differential centrifugation. Mitochondrial protein, calcium and magnesium contents were measured by Lowry method [1]. Beijing Medical University by surface analysis of mitochondrial stereochemical various indicators [2]. The contents of Ca and Mg in the soleus and gastrocnemius muscle of the tail cages were increased compared with the control group on the 15th and 30th days, and all the indexes of stereology were changed to some extent. The results showed that after the tail suspension, the mitochondrial Ca, Mg ion metabolism and stereology of the skeletal muscle both changed, and their changes would inevitably affect the muscle contractile function.
