目的:建立十堰地区连翘药材化学成分的HPLC指纹特征图谱,为连翘药材的质量控制提供科学依据。方法:采用HPLC-梯度洗脱法,以Waters Sun Fire C18(250 mm×4.6 mm,5μm)为色谱柱,Diamonsil C18保护柱,以乙腈和0.4%醋酸溶液为流动相,梯度洗脱,流速为1 ml·min-1,检测波长为277 nm,柱温为30℃。结果:不同产地的12批连翘药材化学成分特征图谱共检出14个共有峰。对不同产地的14个特征峰进行聚类分析,连翘药材产地与特征图谱相似度差异不大,样品中连翘酯苷A和连翘苷的含量有明显差别。结论:该方法简便、快速,可做为连翘药材的质量控制的有效手段。
OBJECTIVE: To establish the HPLC fingerprints of chemical components of Forsythia suspensa medicinal herbs in Shiyan area, and provide a scientific basis for the quality control of Forsythia suspensa medicinal materials. METHODS: HPLC-gradient elution was performed on a Waters Sun Fire C18 (250 mm × 4.6 mm, 5 μm) column and a Diamonsil C18 guard column using acetonitrile and 0.4% acetic acid as mobile phases with a gradient of 1 ml · min-1, detection wavelength was 277 nm, column temperature was 30 ℃. Results: The chemical constituents of 12 batches of Forsythia suspensa from different habitats showed 14 common peaks. Cluster analysis of 14 characteristic peaks in different habitats showed that there was no significant difference in the similarity between the origin and the characteristic map of Forsythia suspense. The contents of Forsythoside A and Forsythin in the samples were significantly different. Conclusion: The method is simple and rapid and can be used as an effective method for the quality control of Forsythia suspensa.
