本文报道了一种质量放大石英晶体微天平免疫分析。抗体包被的石英晶振在待测抗原溶液中孵育时,加入相应的抗体,待测抗原与抗体以复合物的形式结合到晶振上。这种抗原抗体多分子层与通常抗原单分子层结合相比,克服了单分子层结合的抗原分子数较少的限制。应用该方法测定了人血清中的免疫球蛋白(IgM),在10.2~161.0μg/ml范围,响应频率与浓度有较好的线性关系。比较常规的石英晶体微天平免疫传感器方法,灵敏度提高了约3倍。
This article reports a mass-magnification quartz crystal microbalance immunoassay. When the antibody coated quartz crystal is incubated in the antigen solution to be tested, the corresponding antibody is added, and the antigen to be tested binds to the crystal as a complex. This antigen-antibody multi-molecular layer overcomes the less limiting number of antigen molecules bound by the monolayer as compared to the usual antigen monolayer binding. The method was applied to determine the immunoglobulin (IgM) in human serum. There was a good linear relationship between the response frequency and the concentration in the range of 10.2-161.0 μg / ml. More conventional quartz crystal microbalance immune sensor method, the sensitivity increased by about 3 times.
