目的:利用ER+、PR+的人子宫内膜癌细胞株RL-952种植于BALB/c雌性裸鼠形成移植瘤模型后,通过观察移植瘤生长体积的变化、PTEN以及Ki-67的变化来研究来曲唑对子宫内膜癌的治疗效果。方法:将对数生长期的人子宫内膜癌RL-952细胞接种于裸鼠身上形成移植瘤模型,分别给予来曲唑、甲地孕酮、来曲唑+甲地孕酮及生理盐水来对比药物对移植瘤生长的影响。结果:来曲唑组给药后瘤体体积明显小于甲地孕酮组和空白对照组,但与来曲唑+甲地孕酮组差异无统计学意义。PTEN的表达率在来曲唑组、甲地孕酮组、来曲唑+甲地孕酮组均高于空白对照组,Ki-67在各组中的表达无差异。结论:来曲唑对子宫内膜癌移植瘤的抑制作用明显,且与甲地孕酮相比较,优于甲地孕酮。
OBJECTIVE: To study the effects of ER +, PR + on endometrial carcinoma cell line RL-952 implanted into BALB / c female nude mice to establish a xenograft tumor model by observing the changes of tumor volume, PTEN and Ki-67 Therapeutic effect of letrozole on endometrial cancer. Methods: RL-952 cells of human endometrial carcinoma in logarithmic growth phase were inoculated on nude mice to form xenograft tumor model. Letrozole, megestrol, letrozole, megestrol and normal saline Contrast the effect of drugs on the growth of xenografts. Results: The volume of tumor in the letrozole group was significantly smaller than that in the megestrol group and the blank control group, but there was no significant difference between the letrozole group and the letrozole and megestrol group. The expression of PTEN in letrozole, megestrol and letrozole groups was higher than that in blank control group, while there was no difference in the expression of Ki-67 in each group. Conclusion: Letrozole has obvious inhibitory effect on the transplanted tumor of endometrial carcinoma and is superior to megestrol in comparison with megestrol.
