目的:探讨米酒作为引药对瓜蒌薤白体外抗氧化活性及对ISO所致大鼠急性心肌缺血损伤药效学的影响。方法:制备瓜蒌薤白水提物的米酒溶液和水溶液,采用紫外-可见分光光度法测定瓜蒌薤白米酒溶液和水溶液的抗氧化活性。采用ISO制备大鼠急性心肌缺血模型。大鼠随机分为9组,每组8只,正常对照组、模型组分别给予生理盐水,瓜蒌薤白米酒溶液和水溶液按高(5 g/kg)、中(10 g/kg)、低(20 g/kg)剂量给药,复方丹参滴丸组剂量为0.085 g/kg,1次/d,给药7 d,末次给药后,除正常对照组外,各组造模并记录心电图变化,测定血浆及心肌组织NO、T-SOD、GSH-Px、MDA、CAT含量,HE染色观察心肌组织病理学变化。结果:瓜蒌薤白米酒溶液和水溶液的体外抗氧化活性差异无统计学意义。与模型组比较,各给药组大鼠心电图ST段显著降低(P<0.01或P<0.05),血浆及心肌组织NO、T-SOD、GSH-Px、MDA、CAT不同程度改善。与瓜蒌薤白水溶液组比较,瓜蒌薤白米酒溶液组对血浆及心肌组织NO、CAT指标的改善更显著。结论:引药米酒对瓜蒌薤白体外抗氧化活性无显著影响,但可促进瓜蒌薤白体内抗心肌缺血药效的发挥。
Objective: To investigate the effects of rice wine as an anti-oxidant agent on the anti-oxidative activity of melon grubs in vitro and on the pharmacodynamics of acute myocardial ischemia induced by ISO in rats. Methods: The aqueous solution of rice wine and aqueous solution of white water extract from melon grubs were prepared, and the antioxidant activity of white wine liquor and aqueous solution of melon grubs was determined by UV - Vis spectrophotometry. Preparation of rat acute myocardial ischemia model by ISO. The rats were randomly divided into 9 groups with 8 rats in each group. The normal control group and the model group were given normal saline. The white rice wine solution and water solution of the melon grubs were treated with high (5 g / kg), medium (10 g / kg) 20 g / kg). The dose of Fufang Danshen dripping pills group was 0.085 g / kg once a day for 7 days. The histopathological changes of myocardium were observed by HE staining. Results: There was no significant difference in anti-oxidative activity in vitro of melon grubs white rice wine solution and aqueous solution. Compared with the model group, the electrocardiographic ST-segment of the rats in each administration group was significantly decreased (P <0.01 or P <0.05), and the levels of NO, T-SOD, GSH-Px, MDA and CAT in plasma and myocardium were improved to some extent. Compared with the control group, the improvement of NO and CAT index in plasma and myocardial tissue were more significant in the group of white rice wine solution of melon. Conclusion: There is no significant effect on the anti-oxidative activity in vitro of Cuscuta purpurea L., but it can promote the anti-myocardial ischemic effect of Cuscuta purpurea.
