目的:通过观察尿囊素对阿司匹林所致急性胃黏膜损伤大鼠模型胃黏膜血流及细胞因子的影响,初步探讨尿囊素对胃黏膜保护的作用机制。方法:将60只健康雄性Wistar大鼠随机分成阿司匹林损伤组、空白对照组、硫糖铝保护组、氢氧化铝保护组、尿囊素保护组5组。保护组分别提前用硫糖铝、氢氧化铝、复方尿囊素给大鼠灌胃,其后用阿司匹林灌胃致急性胃黏膜损伤,然后分别测定各组大鼠的胃黏膜血流、胃黏膜前列腺素E2(PGE2)及一氧化氮(NO)的含量。结果:各保护组胃黏膜血流与阿司匹林损伤组比较明显增加,尿囊素保护组的胃黏膜血流明显高于氢氧化铝保护组(P=0.000),亦高于硫糖铝保护组,但差异无统计学意义(P=0.073)。尿囊素保护组及硫糖铝保护组胃黏膜PGE2及NO含量均较阿司匹林损伤组明显增加(P=0.001,P=0.000),尿囊素保护组的PGE2及NO含量均明显高于氢氧化铝保护组(P=0.004,P=0.007),亦高于硫糖铝保护组,但差异无统计学意义(P=0.085,P=0.352)。结论:尿囊素可增加阿司匹林致急性胃黏膜损伤大鼠的胃黏膜血流、胃黏膜PGE2及NO的含量,从而发挥胃黏膜保护作用。
OBJECTIVE: To investigate the effect of allantoin on gastric mucosal blood flow and cytokines in acute gastric mucosal injury induced by aspirin in rats, and to explore the mechanism of allantoin on gastric mucosal protection. Methods: Sixty healthy male Wistar rats were randomly divided into 5 groups: aspirin injury group, blank control group, sucralfate protection group, aluminum hydroxide protection group and allantoin protection group. The rats in the protection group were given sucralfate, aluminum hydroxide and compound allantoin in advance, respectively. Afterwards, acute gastric mucosal injury was induced by aspirin, and then the gastric mucosal blood flow, gastric mucosa Prostaglandin E2 (PGE2) and nitric oxide (NO) content. Results: Gastric mucosal blood flow in each protective group was significantly higher than that in aspirin-treated group. Gastric mucosal blood flow in allantoin group was significantly higher than that in aluminum hydroxide group (P = 0.000), and higher than that in sucralfate group But the difference was not statistically significant (P = 0.073). The content of PGE2 and NO in gastric mucosa of allantoin group and sucralfate group were significantly higher than that of aspirin group (P = 0.001, P = 0.000), and the contents of PGE2 and NO in allantoin group were significantly higher than that of hydroxide group The aluminum protection group (P = 0.004, P = 0.007) was also higher than the sucralfate protection group, but the difference was not statistically significant (P = 0.085, P = 0.352). Conclusion: Allantoin can increase gastric mucosal blood flow, gastric mucosal PGE2 and NO content induced by aspirin in acute gastric mucosal lesion, and thus play a protective role in gastric mucosa.
