目的研究茶色素对肾性高血压诱发左室肥厚的预防作用及其可能机制。方法雄性SD大鼠通过部分肾切除造成高血压性左室肥厚模型,分别给予含0.10%和0.25%茶色素的水自由饮用,持续4周。同时设阳性对照组(即模型对照组)和阴性对照组(即假手术组)。测定实验前后大鼠的尾动脉收缩压;实验后大鼠心脏和左室重量及血清AngⅡ浓度(RIA法)。心脏组织中转移生长因子-β(TGF-β)、细胞外调节激酶(ERK)、纤维结合蛋白(FN)、胶原蛋白I(collagen I)等蛋白的表达情况。结果模型对照组大鼠心脏重量(1.71±0.23)g,心脏系数0.42±0.06,左心室重量(1.19±0.22)g,左心室系数0.29±0.06,AngⅡ浓度(748.91±154.80)ng/L。经0.10%和0.25%茶色素处理的大鼠心脏重量分别为(1.54±0.13)g和(1.54±0.14)g,心脏系数分别为0.38±0.04和0.39±0.04,左心室重量分别为(1.07±0.11)g和(1.09±0.11)g,AngⅡ浓度分别为(466.18±111.44)mg/L和(537.55±61.09)ng/L,与阳性对照组比较差异均有统计学意义(P<0.05)。阳性对照组ERK、TGF-β、FN、collagenⅠ蛋白质表达水平较阴性对照组显著增高;0.25%茶色素组ERK、TGF-β、FN、collagenⅠ蛋白质表达水平与阳性对照组比较呈显著降低。结论茶色素能有效预防部分肾切除所引起的左室肥厚,这种作用与茶色素对AngⅡ、TGF-β、FN和collagenⅠ等物质的调节有关,而且茶色素可能对ERK信号转导通路具有一定的抑制作用。
Objective To study the preventive effect of tea pigment on left ventricular hypertrophy induced by renal hypertension and its possible mechanism. Methods Male SD rats were subjected to partial nephrectomy to induce hypertensive left ventricular hypertrophy. The rats were given drinking water containing 0.10% and 0.25% tea pigments freely for 4 weeks respectively. At the same time set positive control group (model control group) and negative control group (ie sham group). The tail artery systolic pressure was measured before and after the experiment. The heart and left ventricular weight and serum Ang Ⅱ concentration (RIA method) were measured. The expression of TGF-β, ERK, FN and collagen I in heart tissue were detected by immunohistochemistry. Results The weight of the model group was 1.71 ± 0.23 g, the cardiac coefficient was 0.42 ± 0.06, the weight of the left ventricle was 1.19 ± 0.22 g, the coefficient of the left ventricle was 0.29 ± 0.06, and the concentration of AngⅡ was (748.91 ± 154.80) ng / L. The heart weights of rats treated with 0.10% and 0.25% tea pigments were (1.54 ± 0.13) g and (1.54 ± 0.14) g, the cardiac coefficients were 0.38 ± 0.04 and 0.39 ± 0.04, the left ventricular weights were (1.07 ± 0.11) g and (1.09 ± 0.11) g respectively, and the AngⅡ concentrations were (466.18 ± 111.44) mg / L and (537.55 ± 61.09) ng / L, respectively. There were significant differences between the two groups (P <0.05). The protein expression levels of ERK, TGF-β, FN and collagenⅠin the positive control group were significantly higher than those in the negative control group. The protein expressions of ERK, TGF-β, FN and collagenⅠwere significantly decreased in 0.25% tea pigment group compared with the positive control group. Conclusions Tea pigment can effectively prevent left ventricular hypertrophy caused by partial nephrectomy. This effect is related to the regulation of tea pigments such as AngⅡ, TGF-β, FN and collagen Ⅰ, and tea pigment may have certain effect on ERK signal transduction pathway Inhibition.
