目的:建立野牡丹主要成分的含量测定方法。方法:样品加入甲醇-25%盐酸溶液(4∶1)混合溶液进行水解,采用RP-HPLC方法测定水解后产生的槲皮素。色谱柱为Shimpack VP-ODS(4.6 mm×150 mm,5μm)C18,以甲醇-0.4%磷酸水溶液(50∶50)为流动相,流速为1.0 mL·min-1,检测波长为360 nm。结果:槲皮素在21.2~169.6μg·mL-1线性关系良好(r=0.999 5),平均回收率为96.3%,RSD 1.54%。结论:方法简便、准确,可靠,可用于野牡丹中槲皮素的含量测定。
Objective: To establish the method for determination of the main components of wild peony. Methods: The sample was hydrolyzed by adding methanol-25% hydrochloric acid solution (4:1), and the quercetin produced after hydrolysis was determined by RP-HPLC method. The chromatographic column was Shimpack VP-ODS (4.6 mm × 150 mm, 5 μm) C18 with a mobile phase of methanol-0.4% phosphoric acid (50:50) at a flow rate of 1.0 mL · min-1 and a detection wavelength of 360 nm. Results: Quercetin showed good linearity (r = 0.999 5) at 21.2-169.6 μg · mL-1, with an average recovery of 96.3% and a RSD of 1.54%. Conclusion: The method is simple, accurate and reliable and can be used for the determination of quercetin in wild peony.
