目的:观察六味地黄汤对糖尿病(DM)大鼠模型肾功能及微小RNA192(microRNA192,mi R-192)、Ⅰ型胶原(collagenⅠ)和转化生长因子-β1(TGF-β1)表达的影响。方法:链脲佐菌素(STZ)腹腔注射法诱导建立大鼠DM模型,模型成功后随机分为3组:模型对照组、卡托普利组、六味地黄汤组,另设正常对照组。治疗8周,观察治疗期间及治疗后大鼠一般状况、血糖、24h尿蛋白(24hUpro)、肌酐、尿素氮、肾质量、相对肾质量(KW/BW),并用实时荧光定量PCR测定肾脏mi R-192、collagenⅠ和TGF-β1mRNA的表达情况。结果:造模3组均出现DM及肾脏损害,模型组肾脏mi R-192、collagenⅠ、TGF-β1mRNA的表达明显增加。各治疗组DM大鼠肾功能改善、24h Upro明显减少(P<0.05,P<0.01),肾质量及KW/BW明显降低(P<0.05,P<0.01),mi R-192、CollagenⅠ、TGF-β1mRNA表达下调;六味地黄汤降血糖的疗效优于卡托普利(P<0.01)。结论:六味地黄汤可以减少mi R-192、collagenⅠ、TGF-β1的产生,延缓糖尿病肾病进程。
Objective: To observe the effect of Liuwei Dihuang decoction on renal function, microRNA192 (mi R-192), collagen Ⅰ and transforming growth factor-β1 (TGF-β1) expression in diabetic rat model. Methods: The model of DM was established by intraperitoneal injection of streptozotocin (STZ). The model was randomly divided into three groups: model control group, captopril group and Liuweidihuang decoction group, and another normal control group. After treatment for 8 weeks, the general condition, blood glucose, 24hUpro, creatinine, urea nitrogen, kidney mass and relative kidney mass (KW / BW) were observed during and after treatment. -192, collagen Ⅰ and TGF-β1 mRNA expression. Results: DM and renal damage occurred in the model group and the expressions of mi R-192, collagen Ⅰ and TGF-β1 mRNA in the model group were significantly increased. The renal function of DM rats in each treatment group was improved, 24h Upro was significantly reduced (P <0.05, P <0.01), renal weight and KW / BW was significantly lower (P <0.05, P <0.01) -β1mRNA expression decreased; Liuweidihuang Decoction hypoglycemic effect is superior to captopril (P <0.01). Conclusion: Liuweidihuang Decoction can reduce the production of mi R-192, collagen Ⅰ, TGF-β1 and delay the progression of diabetic nephropathy.
