目的 :观察脐血、健康人及慢性粒细胞性白血病 (CML )患者外周血 3种不同来源的树突状细胞 (DC)及其抗白血病效应。方法 :分离 3种来源的单个核细胞 (MNC)并在细胞因子 (GM- CSF+IL - 4 +TNF-α)作用下诱导培养 DC。观察 DC形态 ,流式细胞技术检测 DC免疫表型 (CD1 a、CD80 、CD83、HL A- DR) ;将所得 DC经 CML细胞可溶性抗原负载后和其同源 T细胞混合诱导抗原特异性CTL,MTT法检测其杀伤活性。结果 :3种来源 MNC在细胞因子作用下均可获得形态典型的 DC,且高表达 CD1 a、CD80 、CD83、HL A- DR,经抗原负载后的 DC诱导的 CTL 对 CML 靶细胞的杀伤活性也较强。结论 :脐血、健康人及 CML 患者外周血均为理想的 DC来源 ;有较好的诱导 CTL 杀伤白血病细胞效应 ;应用 DC瘤苗免疫治疗白血病必将成为一种崭新的肿瘤免疫治疗方法 ,有十分诱人的发展前景。
Objective: To observe the dendritic cells (DCs) derived from peripheral blood of 3 different types of cord blood, healthy people and patients with chronic myeloid leukemia (CML) and their anti-leukemic effect. Methods: Three kinds of mononuclear cells (MNCs) were isolated and cultured under the action of cytokines (GM - CSF + IL - 4 + TNF - α). The morphology of DCs was observed by flow cytometry, and the immunophenotypes of DCs (CD1a, CD80, CD83, HL-DR) were detected by flow cytometry. The obtained DCs were mixed with their cognate T cells after being loaded by soluble antigen of CML cells to induce antigen- MTT assay to detect its killing activity. Results: MNCs derived from all three kinds of cells could obtain typical DCs under the action of cytokines and high expression of CD1a, CD80, CD83, HL-DR. The killing activity of CTLs induced by DC after antigen loading on CML target cells Also strong. CONCLUSION: Peripheral blood from umbilical cord blood, healthy people and CML patients are the ideal source of DC. CTL can effectively induce the cytotoxic effect of CTL on leukemia cells. Immunotherapy of leukemia with DC vaccine will become a new method of tumor immunotherapy. Very attractive development prospects.
