目的 通过成功纯视细胞移植 ,观察 RCS鼠视皮质内抑制性神经递质 γ-氨基丁酸 (GABA)的分布和变化。 方法 取同龄 Wistar/皇家外科学院 (Royal College of Surgeon,RCS)鼠为供 /受体 ,准分子激光切削法制备视细胞层 ,外路途径移入 RCS鼠的视网膜下腔 ,术后 2周取 RCS鼠手术组眼、手术对照组眼、RCS对照组眼及其视皮质分别做冰冻切片 ,免疫组织化学染色法染色 ,普通光学显微镜下观察。 结果 视细胞移植术后 2周 ,移植视细胞存活 ,外丛状层重建 ,与手术对照眼、RCS对照眼相比 ,重建视网膜受体 RCS鼠对侧视皮质内 GABA免疫反应神经元数量较多 ,而同侧视皮质内无 GABA免疫反应神经元。 结论 成功纯视细胞移植可以重建 RCS鼠的视觉传导通路 ,移植的纯视细胞使 RCS鼠视皮质内 GABA免疫反应的分布发生变化。
Objective To observe the distribution and changes of GABA, an inhibitory neurotransmitter in the visual cortex of RCS rats through successful pure-vision cell transplantation. Methods The Wistar / Royal College of Surgeon (RCS) mice were used as donors / recipients. The retinal layer was prepared by excimer laser cutting. The extracellular route was transplanted into the subretinal space of RCS rats. RCS The rats in the operation group, the operation control group, the RCS control group and the visual cortex were frozen sections, immunohistochemical staining and observation under ordinary light microscope. Results The number of GABA immunoreactive neurons in the cortical repertoire of RCS rats was higher than that of control eyes and RCS control eyes at 2 weeks after transplantation. , While in the same side of the cortex no GABA immune response neurons. CONCLUSIONS: Successful pure-vision cell transplantation can reconstruct the visual conduction pathways in RCS rats. The transplanted pure-vision cells alter the distribution of GABA immunoreactivity in the visual cortex of RCS rats.
