硒代蛋氨酸及亚硒酸钠对小鼠赭曲霉毒素A肾损伤模型的保护作用

目的探讨硒代蛋氨酸、亚硒酸钠干预对小鼠赭曲霉毒素A(OTA)肾损伤模型的保护作用及机制。方法实验分为对照组(A组)、模型OTA组(B组)、硒代蛋氨酸干预组(C组),亚硒酸钠干预组(D组),联合干预组(E组)。采用分光光度法检测小鼠血清和肾组织中丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化物歧化酶(SOD)水平;RT-PCR法检测肾组织谷胱甘肽过氧化物酶(GPx-1、GPx-4)、硫氧还蛋白还原酶(TrxR)mRNA表达,Western bolt法检测肾组织Caspase-3、8、12表达。结果与A组比较,B组GSH-Px、SOD水平降低,MDA水平升高(P<0.01);与B组比较,C、D、E组GSH-Px、SOD水平升高,MDA水平降低(P<0.01);B组GPx-1、GPx-4、TrxR mRNA表达量低于A组,C、D、E组高于B组,E组高于C组、D组(P<0.01);B组Caspase-3、8、12的表达均高于A组,C、D、E组低于B组(P<0.01)。结论硒代蛋氨酸、亚硒酸钠对OTA所致小鼠肾损伤具有保护作用,其机制可能与降低氧化应激反应,上调抗氧化酶表达,减少细胞凋亡有关。 Objective To investigate the protective effect of selenomethionine and sodium selenite on the ochratoxin A (OTA) injury in mice and its mechanism. Methods The experiment was divided into control group (group A), model OTA group (group B), selenomethionine intervention group (group C), sodium selenite intervention group (group D) and combined intervention group (group E). The contents of malondialdehyde (MDA), glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) in serum and kidney of mice were detected by spectrophotometry; The expressions of GPx-1, GPx-4 and TrxR mRNA in renal tissues were detected by Western bolt method. Results Compared with group A, the levels of GSH-Px and SOD were decreased and the level of MDA was increased in group B (P <0.01). Compared with group B, the levels of GSH-Px and SOD were increased and the level of MDA was decreased in groups C, D and E (P <0.01). The mRNA expression of GPx-1, GPx-4 and TrxR in group B was lower than that in group A, and higher in group C, D and E than in group B and group E (P <0.01) The expression of Caspase-3, 8 and 12 in group B was higher than that in group A, but lower in group C, D and E than that in group B (P <0.01). Conclusion Selenomethionine and sodium selenite have protective effects on OTA-induced renal injury in mice. The mechanism may be related to the reduction of oxidative stress, the up-regulation of antioxidant enzymes and the reduction of apoptosis.
