构建基因组文库是分离真核基因,研究基因结构与功能的一个重要手段。为了分离转化生长因子的基因,我们构建了人肺腺癌细胞系LTEP基因组文库。 采用置换型λ噬菌体Charon 28做构建文库的载体,其基因组的非必需片段中两个酶切位点间的DNA片段可以被除去,由外源性DNA片段取代。这种载体具有DNA含量高,体积小,易回收;结构清楚,对插入DNA的顺序分析比较方便;具有较方便地识别重组噬菌体的筛选法;生物学安全等特点。
Constructing a genomic library is an important means of isolating eukaryotic genes and studying the structure and function of genes. To isolate the gene for transforming growth factor, we constructed a human lung adenocarcinoma cell line LTEP genomic library. Using the substitutional lambda phage Charon 28 as a vector to construct a library, DNA fragments between two restriction sites in the non-essential fragments of the genome can be removed and replaced by exogenous DNA fragments. The vector has the characteristics of high DNA content, small volume and easy recovery; the structure is clear, and the sequence analysis of the inserted DNA is convenient; the screening method is more convenient for identifying the recombinant phages; the biological safety and the like.
