目的 :研究肉苁蓉总苷 (Glycosides of Cistanche,GCs)对离体大鼠心脏缺血再灌注损伤的保护作用。方法:采用 L angendorff离体大鼠心脏灌流模型 ,在心脏低灌流期 (5 5 m in) ,给药组在灌流液中分别加入 GCs12 5、2 5 0 mg/ L,以普萘洛尔 (0 .2 μm ol/ L)为阳性对照组 ,以比色法测定心肌 Se- GSH- Px、SOD活性及丙二醛 (MDA)含量 ,电镜检查心肌超微结构的改变。 结果:GCs12 5、2 5 0 mg/ L组明显保护心肌 SOD、Se- GSH - Px活性 ,降低 MDA含量 ,增加再灌后冠脉流量 ,降低冠脉阻力 ,促进心肌收缩力的恢复 ,并明显减轻心肌超微结构损伤。结论:GCs可能是通过清除氧自由基 ,防止脂质过氧化而保护缺血心肌再灌注损伤
Objective: To study the protective effects of Glycosides of Cistanche (GCs) on ischemia-reperfusion injury of rat hearts. METHODS: Using a Langendorff isolated rat heart perfusion model, in the hypoperfusion phase (5 5 min), GCs12 5,250 mg/L were administered to the perfusate in the administration group to propranolol ( 0. 2 μm ol / L) was a positive control group. The activity of myocardial Se-GSH-Px, SOD and malondialdehyde (MDA) were measured by colorimetry. Electron microscopy was used to examine the change of myocardial ultrastructure. RESULTS: GCs12 5 and 250 mg/L significantly protected myocardial SOD and Se-GSH-Px activity, decreased MDA content, increased coronary flow after reperfusion, decreased coronary artery resistance, and promoted recovery of myocardial contractility. Reduce myocardial ultrastructural damage. Conclusion: GCs may protect ischemic reperfusion injury by removing oxygen free radicals and preventing lipid peroxidation.
