目的:研究芹菜素是否能下调CK2α表达和抑制SMMC-7721细胞系肝癌干样细胞自我更新。方法:干细胞培养基超低粘附培养板悬浮培养法扩增肝癌干样细胞;肿瘤球形成法检测肝癌干样细胞的自我更新能力;Western-blot检测细胞CK2α蛋白表达水平。结果:人肝细胞癌SMMC-7721细胞系第2代球细胞具有最强自我更新潜能,可用作肝癌干样细胞模型。芹菜素以浓度依赖方式降低肝癌干样细胞的肿瘤球形成率以及CK2α蛋白表达水平。结论:芹菜素抑制肝癌干样细胞自我更新作用与其下调CK2α表达相关。
OBJECTIVE: To study whether apigenin can down-regulate CK2α expression and inhibit self-renewal of liver cancer stem-like cells in SMMC-7721 cells. Methods: Stem-like cells were expanded by stem cell culture medium suspension culture method. The ability of self-renewal of hepatoma stem-like cells was detected by tumor spheroid formation method. The expression of CK2α protein was detected by Western-blot. Results: Human hepatocellular carcinoma cell line SMMC-7721 has the strongest self-renewal potential and can be used as a hepatoma-like stem cell model. Apigenin reduced the rate of tumor formation and the protein expression of CK2α in HCC-like cells in a concentration-dependent manner. Conclusion: The effect of apigenin on the self-renewal of hepatocellular carcinoma stem cells is related to its down-regulation of CK2α expression.
