目的探讨1,25-二羟维生素D3[1,25(OH)2D3]对Ig A肾病大鼠调节性T细胞的作用。方法 26只Wistar大鼠分为正常对照组、Ig A肾病组、1,25(OH)2D3治疗组。检测大鼠尿蛋白定量(24 h)的变化,采用实时荧光定量聚合酶链反应检测人叉头框蛋白P3(Foxp3)m RNA的表达水平。结果 1Ig A肾病组与正常对照组相比,尿蛋白定量(24 h)水平显著升高,Foxp3 m RNA的水平显著降低,差异有统计学意义(P<0.05)。2治疗组与Ig A肾病组相比,尿蛋白定量(24 h)水平显著降低,Foxp3 m RNA的水平显著升高,差异有统计学意义(P<0.05)。结论11,25(OH)2D3可能降低或缓解Ig A肾病尿蛋白的水平。21,25(OH)2D3可能通过调节调节性T细胞分化发挥免疫作用。
Objective To investigate the effect of 1,25-dihydroxyvitamin D3 [1,25 (OH) 2D3] on regulatory T cells in rats with IgA nephropathy. Methods Twenty - six Wistar rats were divided into normal control group, IgA nephropathy group and 1,25 (OH) 2D3 treatment group. The urinary protein excretion (24 h) was measured by real-time fluorescent quantitative polymerase chain reaction (RT-PCR) to detect the expression of Foxp3 mRNA. Results Compared with the normal control group, the levels of urinary protein (24 h) and Foxp3 mRNA in the IgA nephropathy group were significantly lower than those in the control group (P <0.05). 2 treatment group compared with IgA nephropathy group, urinary protein quantitative (24 h) was significantly lower, Foxp3 m RNA levels were significantly increased, the difference was statistically significant (P <0.05). Conclusion 11,25 (OH) 2D3 may reduce or relieve IgA nephropathy urinary protein levels. 21,25 (OH) 2D3 may play an immunologic role by regulating regulatory T cell differentiation.
