目的 观察纯化的蜂毒肽对兔免疫性关节炎的病理过程是否有抑制作用。方法 采用柱层析方法纯化粗制蜂毒 ,以除去具有溶血及化学刺激作用的杂质。纯化物经CD光谱法和荧光光谱法 ,高压液相色谱分析及氨基酸序列分析。造成家兔卵蛋白性关节炎模型 ,给予蜂毒肽肌肉注射 ,观察组织炎症反应的变化。并测定兔血红蛋白水平 ,进行了体外溶血实验。结果 高压液相色谱分析显示单峰 ,CD光谱法和荧光光谱法、高压液相色谱分析氨基酸序列分析显示纯化物氨基酸序列与文献报道峰毒肽一致。实验组应用纯化蜂毒肽后 ,滑膜炎症明显减轻。溶血实验显示 ,纯化蜂毒肽只有较弱的溶血作用。结论 纯化蜂毒肽对兔免疫性关节炎具有良好的抗炎作用 ,并且溶血作用较弱。
Objective To observe whether purified melittin can inhibit the pathological process of immune arthritis in rabbits. Methods Crude bee venom was purified by column chromatography to remove impurities with hemolysis and chemical stimuli. Purification by CD spectroscopy and fluorescence spectroscopy, high pressure liquid chromatography and amino acid sequence analysis. The rabbit model of ovalbumin arthritis was induced by injection of melittin intramuscularly to observe the changes of tissue inflammatory response. Hemoglobin levels were measured and hemolysis experiments were performed in vitro. Results High-pressure liquid chromatography (HPLC) analysis showed that the amino acid sequence of the purified product was consistent with that reported in the literature by single peak, CD spectroscopy and fluorescence spectroscopy, and high pressure liquid chromatography analysis of amino acid sequence. Experimental group purified melittin, synovial inflammation significantly reduced. Hemolysis experiments show that purified melittin has only weaker hemolysis. Conclusion Purified melittin has good anti-inflammatory effect on immune arthritis rabbits, and hemolytic effect is weak.
