[目的]制备对重症肌无力致病性抗乙酰胆碱受体抗体具有特异性吸附作用的免疫吸附剂.[方法]采用悬浮再生法合成纤维素,以环氧氯丙烷对其进行活化,将人工合成的人乙酰胆碱受体α亚单位67~76位氨基酸为配基,固定于环氧氯丙烷活化后的球型纤维素上,制备出重症肌无力特异性免疫吸附剂,并与乙酰胆碱受体抗体阳性患者的血清进行动态吸附实验,应用酶联免疫吸附法检测反应前后抗体的吸光度,计算抗体吸附率.[结果]以球型纤维素为载体及多肽为配基制备的免疫吸附剂在血清动态吸附实验中对血清中致病性乙酰胆碱受体抗体的吸附率随吸附时间延长而上升,并在吸附开始后2 h时达到高峰,吸附率为26.59%.[结论]以球型纤维素为载体及多肽为配基制备的免疫吸附剂与致病性乙酰胆碱受体抗体吸附性较好,是治疗重症肌无力较理想的免疫吸附剂.
[Objective] The research aimed to prepare immunosorbent for myasthenia gravis pathogenic anti-acetylcholine receptor antibody. [Method] The cellulose was synthesized by suspension-regeneration method and activated by epichlorohydrin. Of human acetylcholine receptor α subunit 67 to 76 amino acids as a ligand, fixed in the epichlorohydrin activated spherical cellulose to prepare a myasthenia gravis specific immunosuppressant and acetylcholine receptor antibody positive The serum of patients was subjected to dynamic adsorption experiments. The absorbance of antibody before and after the reaction was detected by enzyme-linked immunosorbent assay (ELISA) and the rate of antibody adsorption was calculated. [Results] The immunosorbent prepared with spherical cellulose as carrier and polypeptide as ligand In the experiment, the adsorption rate of antibody to pathogenic acetylcholine receptor in serum increased with the prolongation of adsorption time and reached the peak at 2 h after the start of adsorption, with the adsorption rate of 26.59%. [Conclusion] With spherical cellulose as carrier and Peptide ligand for the preparation of immunosuppressants and pathogenic acetylcholine receptor antibody adsorption is better for the treatment of myasthenia gravis ideal immunosorbent.
