目的:动态观察辛热药附子、肉桂、干姜组成的小复方对大鼠物质代谢及第二信使相关指标的影响,以期为虚热证动物模型研究提供实验依据。方法:40只SD雄性大鼠按体重随机分成正常组和中药组,每组20只。中药组大鼠ig附子:肉桂:干姜(1:1:1)200%水煎液16 g.kg-1。在给药12,24,36,48,60 d后眼眶取血2 mL,EDTA抗凝分离血浆,全自动生化仪检测血糖(GLU)、总蛋白(TP)、总胆固醇(TG)、甘油三酯(TC)等物质代谢指标,放免法检测cAMP,cGMP第二信使相关指标。结果:与正常组比较,中药组GLU和TP含量均出现高→低→高的变化趋势,cGMP含量升高;TG,TC,cAMP变化不明显。结论:实验过程中,辛热药小复方对物质代谢和第二信使的影响随着给药时间的延长而出现波动,因此建议在虚热证动物模型研究过程中应根据实验要求选择合适的给药时间。
OBJECTIVE: To dynamically observe the effect of the compound prescription of aconite, cinnamon and ginger on the metabolism of substance and related indicators of second messenger in rats, in order to provide an experimental basis for the study of asthenia syndrome model. Methods: Forty SD male rats were randomly divided into normal group and TCM group according to body weight, with 20 rats in each group. Chinese medicine group rats ig aconite: cinnamon: ginger (1: 1: 1) 200% decoction 16 g.kg-1. Blood samples were collected from orbital eyes at 12, 24, 36, 48 and 60 d after administration, plasma was separated by EDTA anticoagulation, and the contents of GLU, TP, TG, Ester (TC) and other material metabolism indicators, radioimmunoassay detection of cAMP, cGMP second messenger related indicators. Results: Compared with the normal group, the content of GLU and TP in TCM group showed the tendency of high → low → high, cGMP content increased, while the changes of TG, TC and cAMP were not obvious. Conclusion: During the experiment, the influence of the compound prescription of Xin-hot drug on the metabolism of substance and the second messenger fluctuated with the prolongation of administration time.
