欧李果实含有丰富的γ-氨基丁酸,采用超声波提取,结合邻苯二甲醛柱前衍生高效液相色谱法对欧李果实中γ-氨基丁酸的含量进行测定。在对欧李果实提取液柱前衍生后,利用ZORBAX Elipse AAA C18色谱柱,以40mmol/L的磷酸氢二钠(pH 7.8)及V(甲醇)︰V(乙腈)︰V(水)=45︰45︰10,为流动相进行梯度洗脱。以γ-氨基丁酸质量浓度X(μg/mL)对峰面积Y(mAU·min)进行线性回归,得回归方程Y=10.689 1X+6.130 2,相关系数为0.999 6,该方法γ-氨基丁酸的最低检出限为0.064 ng。γ-氨基丁酸平均加标回收率为101.1%,RSD为1.60%。结果证明,该方法具有高效、结果准确、重复性好的特点。通过本法测得欧李果实中γ-氨基丁酸的含量为14.0 mg/100 g。
Oolong fruit is rich in γ-aminobutyric acid, and the content of γ-aminobutyric acid in Oolong plum fruit is determined by ultrasonic extraction combined with phthalaldehyde pre-column derivatization high performance liquid chromatography. The content of γ-aminobutyric acid in the plum fruit was 14.0 mg / 100 g measured by this method.
