使用 ̄3H胸腺嘧啶核苷摄取作为日本血吸虫体外培养DNA合成指标的研究发现:①在体外培养不同时间,血吸虫DNA合成速度不同。在刚进入离体培养的20h内,血吸虫的DNA合成被抑制在较低的水平。随后,血吸虫雌雄虫的DNA合成迅速上升达到最高水平。在培养的1~6d,血吸虫DNA合成虽略有变化,但基本上维持在较高水平。在培养的6~12d,血吸虫DNA合成迅速降低到低水平,②雌雄虫的DNA合成不同,雌虫DNA合成较雄虫高,差别有高度显著性。③培养基中的血清浓度不同,DNA合成速度不同。当低于10%血清浓度,有随着血清浓度的增加.DNA合成也呈增加的趋势。当高于10%血清浓度,随着血清浓度的增加.DNA合成受抑制。在10%血清浓度和20%血清浓度的DNA合成的差别有显著性(P<0.05).证明10%血清浓度是培养基中最适血清浓度。最后,讨论了各种影响体外培养日本血吸虫成虫DNA合成因素的意义。
Using ~ 3H thymidine uptake as an indicator of DNA synthesis in vitro of Schistosoma japonicum found that: ① at different times of culture in vitro, the speed of DNA synthesis of schistosomes is different. Schistosome DNA synthesis was inhibited to a low level within 20 h of in vitro culture. Subsequently, the DNA synthesis of Schistosoma malariae reached its highest level rapidly. In cultured 1 ~ 6d, the DNA synthesis of schistosome although slightly changed, but basically maintained at a high level. In cultured 6 ~ 12d, DNA synthesis of schistosomes rapidly reduced to low levels, ② DNA synthesis of different male and female, female DNA synthesis than male, the difference was highly significant. ③ different serum concentrations in the medium, DNA synthesis rate is different. When less than 10% serum concentration, there is an increase with serum concentration. DNA synthesis also showed an increasing trend. Above 10% serum concentration, with increasing serum concentration. DNA synthesis is inhibited. There was a significant difference in DNA synthesis between 10% serum and 20% serum (P <0.05). Proof that 10% serum concentration is the optimal serum concentration in the medium. Finally, the significance of various factors affecting the DNA synthesis of adult Schistosoma japonicum in vitro was discussed.
