目的:观察龙鳖血清对不同来源干细胞生物特性的影响。方法:从滑液和骨髓中分离培养间充质干细胞(MSCs),以龙鳖血清干预MSCs,观察不同来源MSCs的标记物和炎症因子的表达情况,以及向软骨细胞分化能力的影响。结果:不同来源MSCs培养后的细胞形态及标记物表达无差别,原代SFMSC表达IL-1β、TNF-α和TGF-β1明显高于原代、传代BM-MSC及传代SF-MSC(P<0.01),龙鳖血清可明显抑制SF-MSC分泌炎症因子(P<0.01),而对BM-MSC分泌炎症因子无明显影响;原代SFMSC表达的胶原蛋白1、2和3均低于原代和传代的BM-MSC(P<0.05),龙鳖血清可明显诱导原代SF-MSC和BM-MSC向成软骨细胞表达(P<0.05)。结论:龙鳖血清对SF-MSC和BM-MSC均有抑制细胞因子释放,诱导SF-MSC和BM-MSC向软骨分化的能力,这对中药在防治骨关节炎机理研究中具有重要的指导意义。
OBJECTIVE: To observe the effects of Turtle serum on the biological characteristics of stem cells from different origins. Methods: Mesenchymal stem cells (MSCs) were isolated and cultured from synovial fluid and bone marrow. MSCs were treated with Serum of Turtle Serum and the expression of markers and inflammatory factors of MSCs from different sources were observed. RESULTS: There was no difference in cell morphology and marker expression between MSCs cultured in different sources. The expression of IL-1β, TNF-α and TGF-β1 in primary SFMSC was significantly higher than that in primary, passaged BM-MSCs and passaged SF-MSCs (P < 0.01). Serum of turtle soft-shelled turtle could significantly inhibit the secretion of inflammatory cytokines by SF-MSC (P <0.01), but had no effect on the secretion of inflammatory cytokines by BM-MSCs. The expression of collagen 1, 2 and 3 in primary SFMSC was lower than that of primary And passaged BM-MSC (P <0.05). Serum of Turtle’s soft-shelled turtle could obviously induce the expression of primary SF-MSC and BM-MSCs into chondrocytes (P <0.05). CONCLUSION: Both Tufang turtle serum and SF-MSCs can inhibit the release of cytokines and induce the differentiation of SF-MSCs and BM-MSCs into cartilage, which is of great importance to the prevention and treatment of osteoarthritis .
