为探讨整合素蛋白 β3亚基在胚胎着床期的作用 ,本实验应用特异性抗整合素蛋白 β3抗体 ,采用免疫和 Western印迹法 ,二氨基联苯胺显色 ,VDS扫描等技术 ,检测妊娠 d3、5、7、1 5小鼠子宫内膜及人分泌期子宫内膜和妊娠早期中整合素β3亚基的表达量。结果 :小鼠子宫内膜整合素蛋白 β3亚基表达量在妊娠 d5明显上升 ,至 d7时达高峰 (与对照组比较 ,P<0 .0 1 )。此后下降 ,d1 5时 ,整合素蛋白 β3的表达量与对照组相比 ,无明显差异。人蜕膜组织和分泌期子宫内膜均有整合素蛋白β3的表达。蜕膜整合素蛋白β3的表达量与胚囊大小无关 ,但比分泌期子宫内膜高 4~ 5倍 (P<0 .0 0 0 1 )。这些结果提示 :整合蛋白 β3亚基在子宫内膜中的特异性变化 ,可能启动、促进胚胎的粘附和着床 ,在着床过程中起重要作用。
In order to investigate the function of integrin β3 subunit in embryo implantation, specific anti-integrin β3 antibody was used in this study. Immunohistochemistry and Western blotting, diaminobenzidine colorimetry and VDS scanning were used to detect the expression of d3 , 5,7,1 5 mouse endometrium and human secretory endometrium and early pregnancy expression of integrin β3 subunit. Results: The expression of integrin β3 subunit in mouse endometrium increased significantly at d5 of gestation, reaching its peak at d7 (P <0.01 compared with the control group). Since then, there was no significant difference in the expression of integrin β3 compared with the control group at d1 5. Human decidual tissue and secretory endometrium have integrin protein β3 expression. The expression of decidual integrin β3 was not related to the size of the embryo sac, but it was 4 ~ 5 times higher than that of the secretory endometrium (P <0.01 01). These results suggest that the specific changes of the integrin β3 subunit in the endometrium may initiate and promote the adhesion and implantation of embryos and play an important role in the process of implantation.
