目的探讨ICR小鼠不同发育阶段脑组织原钙黏蛋白7a(protocadherin 7a,Pcdh7a)基因表达及临床意义。方法取胚胎期第16.5、18.5天,出生即刻及出生后第3、7、14、21、30、60天ICR小鼠脑组织,采用实时荧光定量PCR法检测海马和小脑组织Pcdh7a基因表达,采用原位杂交法检测ICR小鼠不同部位脑组织Pcdh7a基因表达。结果小脑组织Pcdh7amRNA相对表达量在胚胎期第18.5天(3.26±0.27),出生即刻及出生后第3、7、14、21、30、60天(4.45±0.23、5.13±0.33、3.47±0.39、5.22±0.75、5.74±0.38、6.48±0.29、6.20±0.29)均明显高于胚胎期第16.5天(1.00±0.03)(P<0.05);胚胎期第16.5、18.5天,出生即刻及出生后第3、7、14、21、30、60天海马组织Pcdh7amRNA相对表达量(1.00±0.04、0.94±0.07、1.06±0.08、0.96±0.08、0.93±0.03、0.82±0.01、0.84±0.01、1.00±0.03、1.00±0.02)比较差异均无统计学意义(P>0.05);原位杂交结果显示,胚胎期第16.5天、出生后第7天Pcdh7amRNA主要在嗅球、海马、皮层和小脑中高表达,出生后第21天主要在海马、小脑、延髓、脑桥高表达,出生后第30天主要在海马、皮层和小脑高表达,且在海马中主要表达于CA1区和DG区。结论 Pcdh7a基因在ICR小鼠不同发育阶段不同部位脑组织呈高表达,可能参与ICR小鼠神经系统发育过程中学习记忆、嗅觉系统及运动功能的调节。
Objective To investigate the gene expression and clinical significance of protocadherin 7a (Pcdh7a) in different developmental stages of ICR mice. Methods The ICD mouse brain tissues immediately before birth and on the 3rd, 7th, 14th, 21st, 30th and 60th day after birth were detected by flow cytometry (FCM). The expression of Pcdh7a in hippocampus and cerebellum was detected by real-time fluorescence quantitative PCR. In situ hybridization was used to detect the expression of Pcdh7a in different tissues of ICR mice. Results The relative expression of Pcdh7amRNA in cerebellum was significantly higher in embryonic day 18.5 (3.26 ± 0.27), immediately after birth and on day 3,7,14,21,30,60 (4.45 ± 0.23,5.13 ± 0.33,3.47 ± 0.39, 5.22 ± 0.75,5.74 ± 0.38,6.48 ± 0.29,6.20 ± 0.29) were significantly higher than those on embryonic day 16.5 (1.00 ± 0.03) (P <0.05). The in situ hybridization results showed that Pcdh7amRNA was mainly expressed in the olfactory bulb, hippocampus, cortex and cerebellum on the 16th day after birth and on the 7th day after birth On day 21, it was mainly expressed in the hippocampus, cerebellum, medulla oblongata and pons. It was mainly expressed in the hippocampus, cortex and cerebellum 30 days after birth and mainly expressed in the CA1 and DG regions in the hippocampus. Conclusions Pcdh7a gene is highly expressed in different parts of ICR mice at different developmental stages and may play a role in the regulation of learning and memory, olfactory system and motor function during the development of nervous system in ICR mice.
