目的:探讨右归丸预防哮喘复发的作用机制。方法:用DNFB诱导小鼠发生迟发性变态反应(DTH)后,评价右归丸的免疫抑制作用;将小鼠切除双侧肾上腺,用二甲苯诱发小鼠耳肿胀,考察右归丸抗炎作用与肾上腺轴的相关性。静脉注射LPS建立急性肺损伤模型,以血清中TNF-α浓度、肺指数、肺脏冲洗液相关指标评价右归丸抗急性肺损伤作用。结果:右归丸20g/kg可显著减少正常小鼠胸腺指数,但对DNFB小鼠耳肿胀、胸腺指数以及脾脏指数均无明显影响。右归丸各剂量组可显著减轻去肾上腺小鼠耳肿胀,右归丸10g/kg可显著减轻LPS小鼠肺指数,右归丸20g/kg可显著降低LPS小鼠血清TNF-α水平,显著降低肺脏组织灌洗液中的单核细胞数量。右归丸有降低肺脏组织NO和TNFα水平的趋势,但作用较弱。结论:提示右归丸防治哮喘可能与其能够减少单核细胞浸润、减少TNFα释放有关,其详细作用机理还有待进一步深入研究。
Objective: To explore the mechanism of You Gui Wan in preventing the recurrence of asthma. Methods: The immunosuppressive effect of Yougui Wan was evaluated after DNT induced delayed hypersensitivity (DTH) in mice. The bilateral adrenal glands were excised and xylene-induced ear swelling was observed in mice. Effect and adrenal axis correlation. The model of acute lung injury was established by intravenous injection of LPS. The effect of Yougui pill on anti-acute lung injury was evaluated by the concentration of TNF-α in serum, lung index and related indicators of lung flushing fluid. Results: Yougui Wan 20g / kg can significantly reduce the normal mouse thymus index, but DNFB mouse ear swelling, thymus index and spleen index had no significant effect. Yougui Wan dose group can significantly reduce ear swelling in adrenal mice, Yougui Wan 10g / kg can significantly reduce the LPS mouse lung index, Yougui Wan 20g / kg can significantly reduce the level of serum TNF-α in LPS mice, significantly Reduce the number of mononuclear cells in lung tissue lavage fluid. Yougui Wan reduce lung tissue NO and TNFα levels trend, but the effect is weak. Conclusion: It is suggested that Yougui Wan can prevent and cure asthma may be related to its ability to reduce infiltration of monocytes and reduce the release of TNFα. The detailed mechanism of action is yet to be further studied.
