目的:通过建立胶原诱导性关节炎(collagen inducted arthritis,CIA)大鼠模型,评价骨碎补总黄酮在抑制CIA大鼠关节骨破坏方面的作用及机制。方法:利用皮下注射牛Ⅱ型胶原诱导Wistar大鼠发病,建立CIA大鼠模型。将造模成功且炎症分值在2分及以上的大鼠进行随机分组并进行相应治疗,评价骨碎补总黄酮对关节骨破坏情况的作用。结果:治疗12周后,骨碎补总黄酮组、骨碎补总黄酮加1/2依那西普组、依那西普组骨小梁面积百分数及骨小梁数量均明显高于灭菌用水组(P<0.05);骨小梁分离度明显小于灭菌用水组(P<0.05)。结论:骨碎补总黄酮能明显抑制CIA大鼠炎症关节骨破坏,其作用可能与骨小梁数量减少及骨小梁分离度的增大相关。
OBJECTIVE: To establish a rat model of collagen-induced arthritis (CIA) to evaluate the role and mechanism of total flavonoids of Rhizoma Drynariae in inhibiting the destruction of joint bone in CIA rats. Methods: The pathogenesis of Wistar rats was induced by subcutaneous injection of bovine type Ⅱ collagen, and CIA rat model was established. The rats with successful modeling and inflammation scores of 2 points and above were randomly divided into groups and treated accordingly to evaluate the effect of total flavonoids of Rhizoma Drynariae on joint bone destruction. Results: After 12 weeks of treatment, the total flavonoids of Rhizoma Drynariae, total flavonoids of Rhizoma Drynariae plus ½ etanercept group, the trabecular area percentage and trabecular number of etanercept group were significantly higher than those of sterilized Water group (P <0.05); trabecular separation was significantly less than the sterile water group (P <0.05). CONCLUSION: Total Flavonoids of Rhizoma Drynariae can significantly inhibit bone destruction of inflammatory joints in CIA rats, which may be related to the decrease of trabecular number and the increase of trabecular separation.
