为了优化三斑海马酶解工艺并对其液化蛋白进行抗氧化研究,本文采用酶法工艺对三斑海马进行液化蛋白的提取,以酶解率为指标,采用了4组单因素实验、16组正交实验进行优化,以DPPH自由基的清除率来衡量三斑海马液化蛋白的抗氧化活性。实验结果表明,三斑海马在使用碱性蛋白酶,料液比1:10、反应时间6h、酶用量1%时提取的液化蛋白水解率最高;三斑海马液化蛋白具有较高的抗氧化活性,对DPPH·的IC50为0.07mg/mL,高于维生素C。
In order to optimize the enzymatic hydrolysis of the three-spot hippocampus and to study the antioxidant activity of the three-spot hippocampus, we extracted the liquefied protein from the three-spot hippocampus using enzymatic technology. Four groups of single- Orthogonal experiments were optimized to DPPH free radical scavenging rate to measure the three spots of hippocampal liquefaction protein antioxidant activity. The experimental results showed that the hydrolysis rate of liquefied protein of the three-spot hippocampus was the highest when alkaline protease was used, the ratio of solid to liquid was 1:10, the reaction time was 6h, and the amount of enzyme was 1%. The three-spot hippocampal liquefaction protein had high antioxidant activity, The IC50 for DPPH was 0.07 mg / mL, higher than vitamin C.
