目的体外实验研究PI3K/Akt抑制剂(LY294002)对鼻咽癌细胞CNE-1放射敏感性的影响。方法体外培养人鼻咽癌细胞CNE-1,本实验分为四组:对照组(A)、单纯放射组(B)、单纯药物组(C)、加药联合放射组(D),按细胞逐步稀释,采用的逐渐增加的放射剂量,分别为0、2、4、6、8、10Gy,PI3K/Akt抑制剂(LY294002)的试验浓度为20mol/L,利用细胞克隆形成实验观察各组CNE-1细胞存活情况,单击多靶数学模型以及L-Q模型拟合剂量存活曲线。结果①LY294002联合照射组Do、Dq、SF2明显比单纯放射组低。CNE-1细胞集落形成实验结果显示:鼻咽癌细胞受不同剂量照射后,B组和D与A组相比,细胞集落形成能力受抑制,放射剂量的增加,集落形成能力受抑制更明显;在相同的放射剂量时,D组细胞集落形成率明显低于B组;根据数据结果在电脑拟合的细胞存活曲线,其结果显示:D组Do值为0.522Gy,Dq值为2.54Gy;B组Do值为0.783Gy,Dq值为2.16Gy,SER为1.36;a/β值B组(1.78Gy)小于D组(9.65Gy)。结论 PI3k/Akt抑制剂(LY294002)联合照射可以提高鼻咽癌细胞CNE-1放射敏感性。
Objective To study the effects of PI3K / Akt inhibitor (LY294002) on radiosensitivity of nasopharyngeal carcinoma cells CNE-1 in vitro. Methods The human nasopharyngeal carcinoma cell line CNE-1 was cultured in vitro. The experiment was divided into four groups: control group (A), radiotherapy alone group (B), simple drug group (C), dosing plus radiotherapy group Gradual dilution, the use of increasing doses of radiation, respectively 0,2,4,6,8,10 Gy, PI3K / Akt inhibitor (LY294002) test concentration of 20mol / L, the use of cell cloning formation experiment observed CNE -1 cell survival, click multi-target mathematical model and LQ model to fit the dose survival curve. The results of CNE-1 cell colony formation assay showed that the colony forming ability of B cells and D cells were inhibited compared with those of A group when the cells were exposed to different doses of radiation. The radiation dose and cell colony formation ability were inhibited more significantly. At the same radiation dose, the cell colony forming rate in group D was significantly lower than that in group B. According to the data fitted in the computer, the cell survival curve showed that the value of Do in group D was 0.522Gy and the value of Dq was 2.54Gy. The Do value was 0.783Gy, Dq was 2.16Gy and SER was 1.36. The value of a / β in group B (1.78Gy) was less than that in group D (9.65Gy). Conclusions The combination of PI3k / Akt inhibitor (LY294002) and radiation can enhance the radiosensitivity of CNE-1 cells in nasopharyngeal carcinoma cells.
